GE HEALTHCARE LIFE SCIENCES / AMERSHAM FRAC-950 is a laboratory instrument designed specifically for fractionation and purification of recombinant proteins from cell culture supernatants. AMERSHAM FRAC-950 is based around two high-pressure pumps, offering up to 4500 psig (312bar) of differential pressure, and a wide range of flow rates, from 0.1ml/min to 10ml/min for buffer and from 1µl/min to 10ml/min for protein-containing solutions. The pumps are accompanied by a wealth of fractionation accessories designed to capture and separate proteins from the culture medium based on different physico-chemical properties. The modular design of the system makes it highly flexible and customisable to laboratory protocols, with the possibility to add or remove features based on the experimental needs. The unit also offers a range of additional capabilities such as online dead-end filtration to remove cellular debris and particulate matter, and ultrafiltration modules with either tangential flow or stirred cell configurations, for removes of macromolecules and cell debris. Other features include isocratic and gradient elution pumps, a range of adjustable and fixed pH isoelectric focusing and an integrated centrifuge. The system can be controlled in either manual or automatic mode, and is equipped with a 21 line display unit for interfacing with the user, allowing for setting of parameters such as flow rate, pressure and temperature, as well as control of the entire process. Furthermore, specialty software can be used to ensure optimization of the fractionation process.
