ILLUMINA MiSeq is a next-generation sequencing (NGS) instrument. It is an automated, fast and simple platform capable of performing high-throughput sequencing with a variety of applications, such as clinical sequencing, gene expression analysis, and de novo sequencing of complex genomes. MiSeq is capable of producing up to 2x300 bp reads with both single-end and paired-end sequencing. ILLUMINA MiSeq is equipped with an ILLUMINA Genome Analyzer IIx sequencing equipment. It uses patterned flow cells to perform sequencing and image-based cluster formation. The flow cell is a chip-like structure made from a specific substrate that enables massively parallel sequencing. It consists of thousands of individually addressable sequencing chemistry channels - termed "clusters". The sequencing chemistry is loaded into each channel before the cluster formation process. The exact amount of chemistry in each cluster is determined by the patterned flow cell, with a different chemistry for each different sequencing chemistry channel. The Illuminina MiSeq system uses sequencing-by-synthesis technology to generate the sequence. It is based on the enzymatic synthesis of the complementary strand of a single stranded DNA (ssDNA) molecule. During the process, a reaction mix, known as a sequencing reaction mix, containing the nucleotides of interest is added to the flow cell. Each nucleotide is then selectively added in its correct position to the ssDNA strand, extending the existing sequence. ILLUMINA MiSeq is a highly automated unit suitable for use in laboratories in academic, clinical and industry settings. It requires minimal setup of only three steps: loading of the flow cell, sample preparation and sequencing run. This machine is incredibly easy to use, with intuitive software controls and on-board electronics that simplify data analysis. MiSeq is an upgrade from the original HiSeq platform, and provides a significant advantage over other existing methods of sequencing. ILLUMINA MiSeq offers longer read lengths than the HiSeq and higher throughput than other instrumentation, such as Sanger and 454 sequencing. Additionally, ILLUMINA sequencing-by-synthesis technology does not require sophisticated hardware, making for a much simpler setup. MiSeq provides a cost-effective and easy to use platform for efficient sequencing in the laboratory.
