APPLIED BIOSYSTEMS / ABI / MDS SCIEX API 4000 is a triple quadrupole (QQQ) mass spectrometer designed for proteomic and quantitative analyses. The equipment consists of a vacuum source, an electrospray ion source, quadrupole mass filter, and a pair of quadrupole analyzers. The vacuum source of the machine supplies a steady stream of inert gas and vacuum pressure as regulated by a turbo-molecular pump. The process of ionization uses an electrospray ionization source which utilizes electrical charges to ionize molecules. The vacuum source then accelerates these ions, allowing the quadrupole mass filter to separate them based on their mass-to-charge ratio (m/z). The quadrupole mass filter is composed of four rods that oscillate at varying frequencies and cause ions of different m/z to travel along different trajectories. The control of the ion trajectories is need to achieve efficient mass/ion selection. The pair of quadrupole analyzers are also connected to the mass filter for monitoring or searching for a targeted selection of ions. After passing through the precursor selection quadrupole, the ions travel along a curved path and reach the second quadrupole of the system, where they undergo fragmentation and undergo analysis. A two-dimensional spectrum is produced, which allows the confirmation of molecular species and the measurement of the relative abundance of all the fragment ions in a gas chromatograph-mass spectrometry unit. ABI API 4000 also features a dual-channel detector, a device that measures the energies and intensities of ions extracted from the mass spectrometer. The device measure energies and intensities of molecules to provide information on nucleic acid sequencing and relative abundance of analytes. MDS SCIEX API 4000 also features advanced software that controls the mass spectrometer and processes the data acquired. The software machine provides flexibility for users to make an array of adjustments to instrument parameters.
